Quality Analysis of Lysionotus pauciflorus from Different Origins by UPLC Coupled with Chemometrics
Objective:To establish the fingerprints of Lysionotus pauciflorus from different habitats by UPLC,and comprehensively analyze the quality of L.pauciflorus by the similarity evaluation(SA)in combination with clustering,principal component analysis(PCA)and other chemo-metrics,so as to provide theoretical basis for its quality evaluation.Methods:The fingerprints of 65 batches of L.pauciflorus were established by UPLC gradient elution,and the similarity evaluation of the UPLC fingerprints of L.pauciflorus medicinal materials was carried out using the traditional Chinese medicine chromatographic fingerprint similarity evaluation system(2012A version).Different analysis,SA,PCA,and orthogonal partial least squares-discriminant analysis(OPLS-DA)were used to comprehensively evaluate the quality of L.pauciflorus.Results:Fifteen chromatographic peaks were selected as the common peaks,and 10 common peaks were identified.The similarity of the 65 batches was 0.316~0.954.PCA analysis screened out five principal components with cumulative variance contribution rate of 79.202%,and obtained the comprehensive score of each batch through the loading of each principal component,reflecting the comprehensive quality.The differential components of L.pauciflorus from Guizhou,Yunnan and Guizhou Provinces were screened out by OPLS-DA as phenylethanoid glycosides and flavonoids.Conclusion:UPLC fingerprint established in this experiment combined with the chemometric evaluation method is simple,feasible and reproducible,and can be used to comprehensively evaluate the quality of L.pauciflorus from different origins.
