Method Validation and Result Analysis of the Paper Method for Total Bacterial Colony in Food
Objective:This thesis aims at exploring the consistency of the paper method of three manufacturers'test pieces in the determination of total bacterial colony in food,carrying out the analysis and contrast of the actual application effects of different manufacturers'test pieces,and providing reference basis for food testing institutions.Method:GB 4789.2-2022 of National Food Safety Standard for Microbiological Examination of Food-Determination of Total Bacterial Colony is used to test the total bacterial colony of three manufacturers'bacterial colony test pieces.The detection of the total number of bacterial colonies in standard bacterial suspensions and artificially contaminated samples is conducted by using the paper method and pouring method,and the statistical analysis is adopted to determine the significance and correlation of the detection results between the two methods.Results:In accordance with the statistical analysis,no significant difference(P>0.05)can be found between the two methods in the detection results of standard bacterial suspension and artificially contaminated samples.The good correlation between the detection results of the two methods is shown(R2>0.996).Conclusion:The results of the total bacterial colony test piece method and the pouring method from the three manufacturers are consistent.The test piece for total bacterial colony is faster and simpler,which is able to reduce the detection process and improve the detection efficiency.Some differences are found to exist between test pieces from different manufacturers,and the laboratory can select appropriate test pieces in line with the sample characteristics.
