正交试验优选壮药龙路通颗粒提取工艺研究

目的:优选壮药龙路通颗粒提取工艺.方法:用高效液相色谱法测定三七皂苷R1,人参皂苷Rg1和Rb1,并以出膏率、三七皂苷 R1,人参皂苷 Rg1 和 Rb1 含量为评价指标.采用单因素法考察加水量、提取时间和提取次数,并通过正交试验优选壮药龙路通颗粒最佳提取工艺.结果:以三七皂苷R1,人参皂苷Rg1和Rb1的含量作为龙路通颗粒质控标准,测得龙路通颗粒中 3 种指标性成分的质量分数分别为三七皂苷R1 45.6~60.6 mg/g,人参皂苷Rg1 159.3~234.4 mg/g,和人参皂苷Rb1 143.8~246.0 mg/g;龙路通颗粒最佳提取工艺为加水煎煮提取3 次,第一次加12 倍量水,第二、三次分别加 10倍量水,每次 1.5 h.结论:优选的提取工艺合理可行,为壮药龙路通颗粒的生产及进一步开发提供了实验依据.
Optimization of Extraction Process of Zhuang Medicine Longlutong Granules by Orthogonal Experiment
Objective:To optimize the extraction process of Zhuang medicine Longlutong granules.Method:The content determination method of Notoginsenoside R1,Ginsenoside Rg1,Ginsenoside Rb1 was established by HPLC.The extraction amount,Notoginsenoside R1 content,Ginsenoside Rg1 content,Ginsenoside Rb1 content were used as evaluation indexes.The single factor method was used to investigate the amount of water added,extraction time,and extraction frequency,and the optimal extraction process for Zhuangyao Longlutong decoction was optimized through orthogonal test.Result:Taking the contents of Notoginsenoside R1,Ginsenoside Rg1,Ginsenoside Rb1 as the quality control standard of Longlutong granules,it was determined that the mass fractions of the three index components in Longlutong granules were Notoginsenoside R1 45.6~60.6 mg/g,Ginsenoside Rg1 159.3~234.4 mg/g,and Ginsenoside Rb1 143.8~246.0 mg/g;The optimal extraction process is to decoct with water three times,adding 12 times the amount of water for the first time,and adding 10 times the amount of water for the second and third times,each time for 1.5 hours.Conclusion:The optimized extraction process is reasonable and feasible,providing experimental basis for the production and further development of Zhuang meidicine Longlutong granules.
