Orthogonal Experimental Design of Ultrasonic Extraction of Chlorogenic Acid in Honeysuckle
Objectiv:In order to obtain the optimum extraction process parameters of chlorogenic acid in honeysuckle.Methods:The effects of two different extraction methods,impregnation method and ultrasonic method,were compared.The single factor test and orthogonal test of ultrasonic extraction of chlorogenic acid in honeysuckle were designed.The concentration of A ethanol(40%,50%,60%),extraction time(20 min,35 min,50 min),ultrasonic temperature(50℃,60℃,80℃)and liquid-solid ratio(20 times,35 times,40 times)were investigated.The effects of different process parameters on the yield of chlorogenic acid in honeysuckle were investigated.Results:The single factor test results of ultrasonic extraction of chlorogenic acid in honeysuckle showed that the best ethanol concentration was 50%,the best time was 35 min,the best temperature was 60℃,and the best liquid-solid ratio was 35 times;the influencing factors of orthogonal test were A>D>C>B,and the best extraction process was A2B2C2D2.Conclusion:The yield of chlorogenic acid obtained by ultrasonic extraction of honeysuckle is higher than that obtained by impregnation of honeysuckle,and the yield is increased by 0.19%.The optimum technological parameters of ultrasonic extraction of chlorogenic acid were as follows:ethanol concentration 50%,extraction time 35 min,ultrasonic extraction temperature 60℃,liquid-solid ratio 35 times.The optimization of the process parameters provides a theoretical basis and data support for the extraction of chlorogenic acid in Guizhou honeysuckle by ultrasonic single factor test and orthogonal test.
