过塘蛇中没食子酸的提取及应用

摘要
过塘蛇是我国民间常用药用植物,本文对过塘蛇中没食子酸的提取方法进行了优化,并对其体外抑菌活性及抗肿瘤细胞活性进行了研究.采用酸提取法,结合正交试验,最佳提取条件为:2.5 mol∙L-1盐酸、料液比1∶75、100℃下酸浸提3.5 h.测得过塘蛇中没食子酸提取量为5.12 mg∙g-1.提取获得的没食子酸,对大肠杆菌、枯草芽孢杆菌和金黄色葡萄球菌均有明显的抑制效果,最低抑菌浓度为51.2 µg∙mL-1.过塘蛇没食子酸对A549和MCF-7肿瘤细胞生长也具有一定的抑制作用,IC50分别为57.33 µg∙mL-1和145.0 µg∙mL-1.
Abstract
Jussiaea repens L.is a commonly used medicinal plant in China.This article optimized the extraction method of gallic acid from Jussiaea repens L.and studied its in vitro antibacterial activity and anti-tumor cell activity.By using the acid extraction method and combining orthogonal experiments,the optimal extraction conditions are:2.5 mol∙L-1 hydrochloric acid,acid extraction at 1:75 and 100℃for 3.5 hours,and the extraction amount of gallic acid from the Jussiaea repens L.is 5.12 mg∙g-1.The extracted gallic acid has significant inhibitory effects on Escherichia coli,Bacillus subtilis and Staphylococcus aureus,with a minimum inhibitory concentration of 51.2 µg∙mL-1.Gallic acid from Jussiaea repens L.also has a certain inhibitory effect on the growth of A549 and MCF-7 tumor cells,with IC50 values of 57.33 µg∙mL-1 and 145.0 µg∙mL-1 respectively.
