Synthesis of PNA Capture Probe and Study on Its Performance in Capturing Lung Cancer EGFR Mutant cfDNA
Peptide nucleic acid(PNA)oligomer is a synthetic DNA analog with a non-natural polyamide backbone.It is widely used in the diagnosis and treatment of genetic diseases due to its strong acid resistance,enzyme resistance and specific hybridization ability.However,cfDNA gene detection for lung cancer is still based on traditional PCR amplification technology combined with gene sequencing technology.The detection time is long and the cost is high,especially for the professional and technical requirements of the detection personnel.In order to solve the above problems,this topic solid-phase synthesis technology was used to prepare PNA capture probe,and rolling circle amplification technology was combined to study the capture performance of PNA probe for lung cancer EGFR mutant cfDNA,thereby exploring new detection methods for tumor cfDNA.Studies have shown that the synthesized PNA capture probe can specifically recognize and capture pM-level EGFR mutant cfDNA.The capture performance meets the sensitivity and accuracy requirements of the detection method,and provides key technology for the establishment of new cfDNA gene detection methods for lung cancer.Provide technical support for cfDNA genetic testing of other diseases.
