基于3D皮肤模型的牡丹籽油抗光老化功效研究

Evaluation for Anti-photo aging Efficacy of Peony Seed Oil by 3D Skin Model

以牡丹籽油为实验材料,利用紫外线(Ultraviolet,UV)照射3D皮肤模型模拟体外光老化环境,通过供试样品牡丹籽油的作用,考察活性氧自由基(Reactive Oxygen Species,ROS)含量、Ⅳ型胶原蛋白(Collagen Ⅳ)表达量及脱氧核糖核酸(Deoxyribonucleic Acid,DNA)损伤指标环丁烷嘧啶二聚体(Cyclobutanepyrimidinedimmer,CPD)阳性细胞率,并进一步评价牡丹籽油对人体皮肤的抗光老化功效.结果显示,牡丹籽油可显著降低UV照射后皮肤模型ROS含量(p<0.01),提升Collagen Ⅳ含量(P<0.01),同时CPD阳性细胞率显著降低(P<0.01).实验表明,牡丹籽油能显著减少经UV照射后皮肤的ROS含量,提升Collagen Ⅳ含量,降低CPD阳性细胞率,从而对皮肤起到一定的抗光老化功效.
Using peony seed oil as experimental material,UV irradiation was used to simulate the in vitro photoaging environment of 3D skin model,and the anti-photoaging efficacy of peony seed oil was verified by detecting the content of Reactive Oxygen Species,the expression of collagen Ⅳ and the positive cell rate of cyclobutanepyrimidinemer,a DNA damage index,through the effect of peony seed oil.The results showed that peony seed oil could significantly reduce the content of ROS(p<0.01)and increase the collagenⅣ content(P<0.01)in skin after UV irradiation,while the CPD positive cell rate decreased significantly(P<0.01).The experiment shows that peony seed oil can significantly reduce the content of ROS,improve the content of collagen Ⅳ in skin after UV irradiation,and reduce the rate of CPD positive cells,thus playing a certain anti-photoaging effect on skin.
