为了研究不同液体培养条件对蝉花虫草生长的影响,以菌丝生长量为考察指标,以碳源、氮源、无机盐、培养温度为考察因素,进行单因素实验和正交实验.在转速为150 r/min,接种量为5%的前提下,采用正交实验的方法最终确定培养基最佳配方为:可溶性淀粉20 g/L,蛋白胨15 g/L,KH2PO4 1 g/L,最佳培养温度为28℃.在此条件下,蝉花虫草菌丝生长量为624.48±6.18 g/L.实验得到蝉花虫草液态培养的最佳培养基配方和培养温度,为蝉花虫草的液态培养条件优化提供了理论基础.
Optimization of Liquid Culture Conditions for Cordyceps cicadae
In order to study the effects of different liquid culture conditions on the growth of Cordyceps cicadae,single factor experiment and orthogonal experiment were carried out with the amount of mycelium growth as the index,carbon source,nitrogen source,inorganic salt and culture temperature as the factors.Under the premise of 150 r/min rotation speed and 5%inoculation amount,orthogonal experiment was used to determine the optimal medium formula:soluble starch 20 g/L,peptone 15 g/L,KH2PO4 1 g/L,and the optimal culture temperature was 28℃.Under these conditions,the mycelial biomass of Cordyceps cicadae was 624.48±6.18 g/L.The optimum culture medium formula and culture temperature were obtained,which provided a theoretical basis for the optimization of liquid culture conditions of Cordyceps cicadae.
