Simultaneous Determination of Five Components in Fengre Ganmao Granules by HPLC
Objective:Establish a method for simultaneously determining the content of 5 components in Fengre Ganmao granules.s.Methods:Agilent Eclipse Plus C18 chromatographic column(250 mm×4.6 mm,5 μm)Set the column temperature to 25℃;Gradient elution with acetonitrile 0.1%phosphoric acid solution as mobile phase at a flow rate of 1.0 mL/min;Multi wavelength conversion measurement,detection wavelength:327 nm,278 nm;Conduct methodological validation on the content determination methods of chlorogenic acid,burdock glycoside,isochlorogenic acid A,forsythrin,and rutin,Evaluate the experimental results using cluster analysis(CA).Results:The five components in Fengre Ganmao Granules showed good linear relationships within their respective concentration ranges(n≥0.9991),with average sample recovery rates of 98.67%,97.44%,97.94%,98.44%,100.03%,and RSD of 1.61%,1.38%,1.70%,1.55%,and 0.96%,respectively;The contents of chlorogenic acid,rutin,isochlorogenic acid A,forsythrin,and arctiin in 10 batches of samples were 0.091 mg/g~0.227 mg/g,0.094 mg/g~0.258 mg/g,0.084 mg/g~0.274 mg/g,0.080 mg/g~0.217 mg/g,and 0.809 mg/g~2.584 mg/g;10 batches of samples can be clustered into two categories,s3,s10,s4,s2,s5,and s9 can be clustered into one category,and s1,s8,s6,and s7 can be clustered into one category.Conclusion:The established method is scientific and accurate,and can be used for the content determination of Fengre Ganmao granules.
