Plasma Protein Binding Rate of Olaparib in Rat Plasma by HPLC
The plasma protein binding rate of olaparib in rat plasma was determined by the high-performance liquid chromatography method,and the plasma protein binding rate of olaparib was determined by equilibrium dialysis.The plasma protein binding ratio of three concentrations of olaparib was measured using Kromasil Column C18 column with a mobile phase of methanol:water(50∶50),the detection wavelength of 275 nm,and equilibrium dialysis combined with HPLC-UV method.At low,medium and high concentrations,plasma protein binding to rats was 80.09%,78.44%,and 79.06%,respectively,with a mean value of 79.20%.The results show that the plasma protein binding rate of olaparib can be measured accurately and quickly measured by HPLC and equilibrium dialysis.
