Objective:To study the the chemical constituents of coumarins from the branches and leaves in Citrus medica L.var.sarcodactylis Swingle. Methods:The separation and purification of the chemical components of Citrus medica branches and leaves were carried out by solvent extraction, extraction, macroporous resin column chromatography, silica gel chromatography, gel chromatography,thin-layer chromatography, and preparative high-performance liquid chromatography. The structures of the compounds were characterized by physicochemical properties,1H-NMR and 13C-NMR spectroscopy. Results:Eight coumarins were isolated from the branches and leaves of Citrus medica.Their structures were identified as Scopoletin(1),Oxypeucedanin hydrate(2),2'-Methoxyproarlin hydrate(3),5,7-Dimethoxycoumarin(4),Bergapten(5),5-Hydroxy-7-methoxycoumarin(6),7-Hydroxy-5-methoxycoumarin(7),7-[[(2E,6R)-6,7-Dihydroxy-3,7-dimethyl-2-octen-1-yl]oxy]-2H-1-benzopyran-2-one(8).Conclusion:Among the eight coumarins, compounds 1,2 and 3 were isolated from the leaves of Citrus medica for the first time, and compounds 6,7 and 8 were isolated from the branches of Citrus medica for the first time,it can provide a scientific basis for the analysis of coumarins in Citrus medica.
