Investigating the Effects of Different Factors on the Extraction of Black Soldier Fly(Hermetia illucens L.)Protein by Alkaline Extraction Method based on Subcritical Extraction Assistance
In this study,the black soldier fly(Hermetia illucens L.)larva powder was defatted by subcritical extraction technique,and the protein extraction of defatted black soldier fly larva powder was carried out by four methods,and the optimal method was determined and then further explored the optimal level of the optimal extraction method under various factors.The subcritical extraction technique used butane as solvent,temperature 45 ˚C,pressure 0.4 MPa,40 min/time,and extraction 3 times to obtain the defatted black soldier fly larva powder with 87.09%defatted rate.The protein extraction rates of the four extraction methods were as follows:alkaline extraction method(53.17±2.97%),trypsin extraction method(5.40±0.36%),salt extraction method(5.13±0.84%),and Tris-HCl buffer extraction method(7.27±0.40%).To explore the optimum level of alkaline extraction method under four factors,the results of the one-way test were the concentration of NaOH solution(25 g·L-1),the extraction time(1.5 h),the temperature of the extraction(60 ˚C),and the liquid-feed ratio(25 mL·g-1).
