Optimization Conditions of Solid Fermentation of White Rot Fungi
Objective:Laccase is a copper containing polyphenol oxidase that can catalyze the oxidation of various phenolic compounds and aromatic amines.It has broad application prospects in fields such as food,papermaking,energy,and polymer synthesis.This study focuses on a white rot fungus and uses crop straw as raw material for solid-state fermentation to optimize its laccase production conditions,providing technical support for better utilization of this fungal resource.Methods:Using a single factor experiment,the conditions for solid-state fermentation of white rot fungi to produce laccase were optimized from four aspects:fermentation time,solid-liquid ratio,temperature,pH,etc.Results:When the culture medium is corn cob and bran,with an inoculation amount of 10%,fermentation time of 4 days,solid-liquid ratio of 1∶2.5,fermentation temperature of 28℃,and pH of 5.5,the strongest laccase activity is produced,with a laccase activity of 98.99 U/g.Conclusion:This white rot fungus has a strong ability to utilize crop straw to produce laccase,which can provide a theoretical basis for its comprehensive utilization of biomass energy such as crop straw.
