基于PIK-75探究AEP对细胞骨架的影响

天冬酰胺内肽酶(asparaginyl endopeptidase,AEP)在肿瘤细胞中高表达并发挥至关重要的作用,PIK-75 作为一种抑制剂,它能够诱导凋亡,在治疗肿瘤方面存在一定优势,我们在给予肿瘤细胞PIK-75 后,通过Western Blot实验验证了AEP的蛋白含量的变化,由于细胞骨架在肿瘤增殖迁移中发挥的关键作用,我们在 AEP 蛋白量下降的基础上对肿瘤细胞进行细胞骨架的免疫荧光染色,验证PIK-75 在降低了AEP蛋白量的情况下对细胞骨架形态的影响。
The Effect of AEP on Cytoskeleton was Investigated based on PIK-75
Asparaginyl endopeptidase(AEP)is highly expressed in tumor cells and plays a crucial role.PIK-75,as an inhibitor,is capable of inducing apoptosis and demonstrates certain advantages in the treatment of tumors.After administering PIK-75 to tumor cells,we verified the alteration in the protein content of AEP via the Western Blot experiment.Given the key role of the cytoskeleton in tumor proliferation and migration,on the basis of the decreased protein quantity of AEP,we conducted immunofluorescence staining of the cytoskeleton on tumor cells to validate the impact of PIK-75 on the morphology of the cytoskeleton when the amount of AEP protein was reduced.
