Biochemical Functional Analysis of Key Enzyme for the Synthesis of Polyketene Heteroterpenoids from Deep-sea Fungus Phomopsis tersa FS441
Phomeroids are new skeleton compounds of polyketene heteroterpenoids with excellent anti-tumor activity,which are isolated from marine fungus P.tersa FS441.It is predicted that the ctg1509 gene encoding terpenoid cyclases 1509TC is responsible for the biosynthesis of phomeroids.The ctg1509 gene was expressed and in Escherichia coli and 1509TC protein was purified by affinity chromatography,the in vitro biochemical function of purified protein 1509TC were analyzed.The results indicate that the 1509 TC protein has 336 amino acid residues with a relative molecular weight of 38539.86 Da.It is a novel acidic hydrophilic terpenoid cyclase protein.1509TC can recognize farnesyl pyrophosphate as a substrate and convert it into a new product.This study preliminarily validated the in vitro biochemical function of 1509TC,laying a biochemical basis for the excavation of diverse phomeroids.
