Study on the Separation of Saccharum Alhagi Oligosaccharides by Ultrafiltration and Structural Identification
In this study,the Saccharum Alhagi medicinal herb was screened for impurities and extracted with hot water.The extract was sequentially separated through a 0.16 µm microfiltration membrane and an ultrafiltration membrane with a molecular cut off 10000 Daltons.The filtrate passed through the 10000 Daltons ultrafiltration membrane was concentrated to obtain the Saccharum Alhagi oligosaccharide fraction,with a yield of 70.12%;This part was purified by ion-exchange resin and dextran gel chromatography column to obtain highly purified oligosaccharides with a yield of 55.79%and a sugar content of 92.87%.Through monosaccharide analysis,methylation,and mass spectrometry analysis,the purified Saccharum Alhagi oligosaccharides obtained by this method were identified as trehalose.The purity of trehalose obtained from this research method is high,and structural analysis of trehalose oligosaccharides is carried out,which can provide reference for the application and development of trehalose oligosaccharides in functional food,medicine,and cosmetics.
