红心杉组培苗增殖培养基的优化

Optimization of culture medium for plantlets of Cunninghamia lanceolata (Lamb.) Hook.in tissue culture proliferation

以优良红心杉〔Cunninghamia lanceolata(Lamb.)Hook.〕为材料,采用L9(34)正交试验设计,研究基本培养基、细胞分裂素苯基噻二唑基脲(TDZ)及香蕉汁浓度对其组培苗增殖系数、叶绿素含量、可溶性糖含量的影响.结果表明:对红心杉组培苗增殖系数的影响大小表现为基本培养基>香蕉汁浓度>TDZ,对叶绿素和可溶性糖含量的影响大小表现为基本培养基>TDZ>香蕉汁浓度.经坐标综合评定发现,3/4MS+香蕉汁100 g/L为红心杉组培苗增殖培养的最佳组合,增殖系数可达2.45、叶绿素含量达2.45 mg/g、可溶性糖含量达33.47 mg/g.
Using Cunninghamia lanceolata (Lamb.) Hook as material,through L9(34) orthogonal test design,the effects of MS medium,TDZ concentration and banana juice concentration on the proliferation coefficient,chlorophyll content and soluble sugar content of plantlets were studied. The results showed that:The effects on the multiplication coefficient of the plantlets in tissue culture were as follows:basic medium > banana juice concentration> TDZ concentration. The effect on the content of chlorophyll and soluble sugar was as follows:basic medium > TDZ concentration > banana juice concentration. It is found by comprehensive evaluation of coordinates that,3/4MS + banana juice 100 g/L was the best combination for the multiplication and culture of the plantlets in tissue culture. Under this condition,the proliferationg coefficient was 2.45,the chlorophyll content was 2.45 mg/g, the soluble sugar content was 33.47 mg/g.
