Effects of Polysaccharides of Atractylodes Macrocephala on Antioxidant Capacity and Apoptosis in the Spleen of Goslings Treated with LPS
Investigate the effects of polysaccharides of Atractylodes macrocephala(PAMK)on the antioxidant capacity and apoptosis of gosling spleen after different times of LPS treatment.Eighty 1-day-old Magang geese were randomly divided into four groups,and the diet was supplemented with PAMK.2 mg/kg LPS was injected intraperitoneally on the 24th,26th and 28th day of age and spleen was collected 1 hour after the injection.The pathological changes of spleen tissue were observed.The mRNA expressions of spleen antioxidant enzymes(CAT,GSH-Px,SOD),apoptotic factors(Apaf-1,Caspase3,caspase9 and FAS)and anti-apoptotic factor BCL2 were detected by RT-PCR.PAMK can significantly protect the spleen of goslings from the tissue damage caused by LPS,and has a good anti-oxidation and alleviating the apoptosis of spleen cells effect.Among them,PAMK can restore antioxidant capacity of spleen treated by LPS,while PAMK has a certain lag effect on SOD.In addition,PAMK significantly increased the anti-apoptotic ability of spleen after LPS treatment 1,2 and 3 times,and the anti-apoptotic effect was most obvious after LPS treatment 3 times.
