Analyzsis of Population Genetic Structure of Specific Pathogen Free New Zealand Rabbit by Short Tandem Repeat Scanning Technology
The STR scanning technology was used to conduct genetic detection and analysis on SPF New Zealand rabbits in Guangdong Medical Experimental Animal Center,so as to master the genetic structure of the rabbit population,provide basis for formulating breeding plans,and provide reference for rabbit genetic analysis methods.The venous blood from the ears of 33 rabbits was collected to extract genomic DNA.23 microsatellite loci with rich polymorphism were selected for PCR amplification of genomic DNA.The amplified products were done by STR scanning.The scanning results were done genetic structure analysis of the population include observe the number of alleles,effective alleles,vanillin index and effective heterozygosity by popgene32 software.The results showed that the PCR amplification of all 23 microsatellite loci were well,the average observed allele of the rabbit population is 1.5217,the average effective allele count is 1.2786,the average Shannon index is 0.2442,and the average effective heterozygosity is 0.1564.The genetic structure of the SPF New Zealand rabbit population was preliminarily mastered.The results indicated that the STR scanning technique and the 23 microsatellite loci selected were suitable for the detection and analysis of the genetic structure of rabbit population,and would provide reference and basis for the rabbit genetic analysis methods.
