Expression Analysis of Goose TLR4 and Relevant Immune Factors in Different Tissues Induced by Avian Influenza Vaccine H5 Subtype
In order to make a profound study on expressive characteristics of TLR4 and associated immune factors during poultry vaccination.We used the Sanhua goose as experimental animal to analyze TLR4 and associated immune factors level in different tissues by real-time PCR method and immunohistochemistry technique on 24h after injection of H5 subtype avian influenza vaccine(AIV).The results showed that TLR4 mRNA was higher than the control group in liver and bone marrow(P<0.05),It was significantly higher in spleen,bursa of Fabricius and thymus than that in control group(P<0.01).IL-1β,IFN-α and IFN-γ mRNA level are significantly higher than those of the control group in liver and thymus(P<0.01),IL-6 and IFN-α with significant differences in spleen(P<0.01),but IL-1β mRNA level,were higher than control group in spleen and bursa(P<0.05).The immunohistochemical test indicated that TLR4 protein levels was significant higher than control group(P<0.01),and higher than in liver(P<0.05),which was consistent with the transcriptional level.It suggested that TLR4 and associated immune factors might play roles by injection H5 subtype AIV.
