The Cell Culture of Chicken Intestinal Epithelial Cell Based on Feeder Layer Cells and Effects of Echinacea Extract on cell Proliferation
In order to explore the possibility of using chicken intestinal fibroblasts as feeder cells to cultivate chicken intestinal epithelial cells and the proliferation effect of Echinacea extract on IEC.In this experiment,the chicken intestinal fibroblasts were obtained by digestion by the type IV collagenase,and were prepared into feeder cells using mitomycin C and co-cultured with isolated chicken intestinal epithelial cells.The expression of chicken IEC Cytokeratin protein was determined by cellular immunofluorescence,and the proliferation ability of chicken cells and the effect of echinacea extract on the proliferation were detected by CCK-8 method.The isolated chicken intestinal epithelial cells expressed Cytokeratin protein,had good proliferation ability in co-culture with feeder cells,and survived for 14 days in vitro.In addition,the damaged chicken intestinal epithelial cells could promoted effectively by echinacea extract of 31.25-2000 μg/mL The results show that chicken intestinal fibroblasts can be used as feeder cells to culture chicken intestinal epithelial cells,and echinacea extract can promote their proliferation.This experiment provides a new way for IEC culture in vitro,and also provides a reference for the study of echinacea extract on intestinal mucosal immunity.
