Correlation analysis between Polymorphic Sites of the ACAN gene,Growth and Carcass Traits in Geese
The aggrecan gene(ACAN)is an important candidate gene influencing animal growth traits,and the aggrecan encoded by ACAN is crucial for the structural formation and development of cartilage and bone.In this study,Shitou geese,Magang geese,and Wuzong geese were used as experimental materials.A mixed DNA pool combined with sequencing was employed to screen polymorphic sites in the exon region of the ACAN.The genotypes of SNP polymorphic sites were identified using PCR-RFLP and sequencing of PCR products.Association analyses were then conducted between these SNPs and growth traits in Wuzong,Magang,and Shitou geese,as well as carcass traits in Shitou geese.A total of seven SNP polymorphic sites were identified in the exon region of the ACAN across the three goose populations in this study.Among them,the polymorphic sites g.29647T>C,g.31095C>T,and g.30246C>T were highly significantly associated with breast depth,while g.30135A>G was significantly associated with breast depth.The polymorphic sites g.29647T>C,g.30135A>G,and g.30246C>T showed significantly associated with semi-eviscerated carcass weight,leg muscle weight,and foot weight in Shitou geese.In particular,g.30246C>T was highly significantly associated with foot weight,and g.30085A>G was significantly associated with foot weight.This study can provide some reference for molecular marker assisted breeding of geese.
