Fabrication and Properties of Highly Aligned Electrospun Collagen Fibers
Electrospun collagen fibers(ECF)are fabricated,and the effects of electrospun solution composition,electrospun conditions and environmental factors on electrospun collagen fibers are systematically studied in this article.Scanning electron microscopy is used to analyze the morphology of ECF.The results show that ECF with uniform surface morphology can be obtained under a wide range of electrospun conditions:70%-80%weight concentration acetic acid solution as solvent,collagen weight concentration as 15%-18%,injection speed of electrospun solution as 0.15-0.3 mL/h,electrospun voltage as 16-24 kV,spinning gap as 8-12 cm,22G or 21G blunt needle as spinning nozzle,the relative humidity as 50%-55%,and temperature at 30-32 ℃ of electrospun chamber.Based on the above conditions,aligned electrospun collagen fibers(AECF)are prepared by the disc-like directional fiber collector.Average angle between fibers and Herman orientation factor are used to characterize fiber orientation,and it is found that AECF orientation increased with collector speed.The triple helix structure and self-assembly capacity of collagen in AECF are characterized by circular dichroism and transmission electron microscopy respectively,while the cytotoxicity of AECF is evaluated by Thiazolgl Blue(MTT).The results show that collagen in AECF has a natural collagen-like triple helix structure,self-assembly ability,no cytotoxicity,making it suitable for tissue engineering scaffolds.
