Preparation of In-situ Rapid Gelation Dialdehyde Starch/Collagen Hydrogel with Coagulation Effect
In order to improve the gelation speed of collagen hydrogel at room temperature,dialdehyde starch/collagen(DAS-Col)hydrogel with coagulation function was prepared based on DAS and pigskin collagen.The effects of DAS aldehyde group content and dosage,collagen molecular dosage,Ca2+concentration and pH value on gelation time were investigated.The structure,Young's modulus,coagulation index in vitro(BCI)and total coagulation time of the hydrogel were characterized.When a 10 g/L DAS solution with an aldehyde content of 33.48%and a 10 g/L collagen solution were mixed in a volume ratio of 1∶4,Ca2+concentration was 10 g/L,and the pH value was 7.4,the DAS-Col hydrogel was formed rapidly at room temperature,the gelation time was as short as 93 s,the Young's modulus reached 143.1 kPa,the BCI was 0.106,and the total coagulation time in vitro was 152 s.Moreover,the DAS-Col hydrogel also exhibited excellent self-degradation and self-healing properties.The cross-linking of the DAS with huge branched chains and the collagen molecules would form a huge and staggered hydrophilic hydrogel network skeleton,which was the key to the rapid formation of hydrogel at room temperature.All the research results indicate that DAS-Col hydrogel will be a kind of hydrogel that can be rapidly gelled in situ,and it has the potential to be used for hemostasis care of some irregular wounds on the body surface and deep wounds in the body.
