新型奥沙利铂类抗癌药物(LLC-202)的合成、表征及稳定性

采用氯亚铂酸钾作为起始原料,引入胆酸作为辅助配体,成功合成了一种新型含铂配合物,命名为LLC-202.对该化合物的化学结构进行了全面的表征,包括元素分析、红外光谱(FTIR)和核磁共振波谱(NMR)分析、质谱等.表征结果证实LLC-202符合目标化合物的结构.进一步考察该化合物的稳定性,结果表明其水溶液在37℃时,72h内稳定;在水中会发生缓慢的水合反应,生成有生物活性的药效基团[A2Pt(H2O)2]2+.
Abstract
A novel platinum-containing complex,named LLC-202,was successfully synthesized by using potassium chloroplatinate as the starting material and introducing cholic acid as the auxiliary ligand.The chemical structure of the compound was comprehensively characterized by elemental analysis,infrared spectroscopy(FTIR)and nuclear magnetic resonance spectroscopy(NMR)analysis,mass spectrometry and so on.The characterization results confirmed that LLC-202 conforms to the structure of the target compound.The stability of the compound was further investigated,and the results showed that its aqueous solution was stable for 72 h at 37 ℃;it would undergo a slow hydration reaction in water to produce the biologically active pharmacophore[A2Pt(H2O)2]2+.
