The Regulatory Effect of Compound Xinshuaikang Capsule on the Expression of SERCA2a in Rats with Heart Failure
Objective To observe the effect of compound Xinshuaikang capsule on the expression of SERCA2a and cardiac function in rats with heart failure.Methods A total of 120 male SD rats with SPF grade were selected and divided into blank group(n=20)and model group(n=100).The rat heart failure model was constructed by adriamycin intraperitoneal injection.After successful modeling,the rats(n=91)were randomly divided into model group(n=19),digoxin group(n=18),compound Xinshuaikang low-dose group(n=18),medium-dose group(n=18)and high-dose group(n=18).Cardiac function indexes,SERCA2a and its mRNA gene expression levels were detected in each experimental group.Results Compared with blank group,the expression levels of LVEDD,LVFS,NT-proBNP and miR-25 were significantly increased in model group,digoxin group and Compound Xinshuaikang low-middle-high dose group,while the expression levels of SERCA2a,its mRNA and LVEF were significantly decreased(P<0.05).Compared with model group,the expression levels of LVEDD,LVFS,NT-proBNP and miR-25 were significantly decreased in compound Xinshuaikang low-medium-high-dose group and digoxin group,while SERCA2a and its mRNA expression levels and LVEF were significantly increased(P<0.05).Conclusion Compound Xin-shuaikang capsule can improve the cardiac function of CHF rats by inhibiting the expression of SERCA2a related protein by inhibiting miR-25.
