Simultaneous Analysis of Multiple Chemical Components in Rongjin Zhuanggu Prescription by High Performance Liquid Chromatography
Objective To establish a method for the extraction and qualitative and quantitative analysis of Rongjin Zhuanggu prescription by high performance liquid chromatography(HPLC).Methods The extraction effect of methanol,ethanol,pure water and n-butanol on the lyophilised powder of the aqueous extract of Rongshi Zhuanggu prescription was investigated on an Agilent EC-C18(4.6 mm×250 mm,5 μm)column with the mobile phase of acetonitrile-water in a gradient elution mode with the column temperature of 30℃,the flow rate of 1 ml/min,and the detection wavelength of 212 nm,and the stability of the method was evaluated,and the content of several substances in the compound was finally detected.The contents of several substances in the compound were finally detected.Results Under the optimal chro-matographic conditions,the separation effect of various chemical components in Rongjin Zhuanggu prescription was good,and it was suitable for the quantitative analysis of five components in Rongjin Zhuanggu prescription as follows:catalpa,paeoniflorin,paracosaponin Ⅵ,psor-alen and isopsoralen.The content of catalpol,paeoniflorin,paeoniflorin Ⅵ,psoralen and isopsoralen were 38.30 mg/g,8.13 mg/g,30.18 mg/g,3.89 mg/g and 2.02 mg/g respectively in Rongjin Zhuanggu prescription.Conclusion The HPLC method established in this study has the advantages of high recovery and good reproducibility,and is suitable for the qualitative and quantitative analysis of various compo-nents in Rongjin Zhuanggu prescription.
