A Comprehensive Experimental Design for the Identification and Determination of Isopentenyl Flavonoids from Epimedium Brevicornum
In order to deepen the students'understanding of the active components isopentenyl flavone from Epimedium brevicornu,this comprehensive experiment was designed.The total extract was obtained by distillation and concentration,which was dissolved in water and extracted successively with petroleum ether,ethyl acetate and n-butanol,and then gave petroleum ether phase,ethyl acetate phase and n-butanol phase.The three phases were sampled separately on thin-layer chromatography plates,and their fluorescence color and intensity were observed at 254 nm and 365 nm under UV lamp,and then the spraying sulphuric acid and spraying ferric chloride on sampled thin-layer chromatography plates,which indicated contain flavonoids.The content of isopentenyl flavone in the petroleum ether,ethyl acetate and n-butanol phases were determined by standard curve and UV spectrum,and the repeatability of the method and the stability of the sample were also investigated.
