ISSR标记在中国野生葡萄分类中的应用

利用ISSR标记对包含16个中国葡萄野生种及其近缘植物的81份材料进行分类研究,从100条引物中筛选出13条带型清晰、重复性高、多态性好的引物用于ISSR扩增,共扩增出分子量在300~2000 bp的185条带,其中多态性条带175条,多态百分率为94.6%.根据ISSRR扩增结果,利用NTSYSpc2.10e软件进行相似性系数分析,81份葡萄种质的遗传相似性系数为0.57-0.99.通过UPGMA进行聚类分析,在遗传相似系数为0.83处,79份真葡萄亚属材料聚成22类,聚类结果与传统的分类结果基本一致.同时,根据ISSR聚类结果,将供试材料毛葡萄1099确定为桑叶葡萄,对燕山葡萄0947是否为燕山葡萄提出质疑,并检测出供试的变叶葡萄材料不是一个纯种,而是存在种间的过渡类型.
Application of ISSR markers in taxonomy of Chinese wild grape
16 Chinese wild grape species and their relatives were studied by ISSR markers on taxonomy.13 primers selected from 100 primers were used for ISSR amplification. 185 bands were generated which molecular weight ranged from 300 to 2 000 bp, of which 175 bands were polymorphic bands (the percentage of polymorphic band, PPB=94.6%). According to the results of ISSR amplification, the similarity coefficient were analyzed by NTSYSpc2.10e software, the genetic similarity coefficient was 0.57 to 0.99. The clustering dendrogram was constructed by UPGMA method to explore the taxonomic relationship between 81 Vitis materials. To employ Ampelopsis sinica (Miq.) W. T. Wang. and Vitis rotundifolia as Outgroup,which had genetic similarity coefficient 0.57 and 0.60 with Euvitis, respectively. The 79 Euvitis materials were classified as 22 groups according to the cluster analysis of the PCR amplified products. Most of materials had the same traditional classification but some not. According to the results of ISSR clustering ,we determined the Maoputao 1099 as Vitis ftcifolia Bunge. Whether Yanshanputao 0947 is Vitis yeshanensis J. X. Chen was suspected. We also found inter-specie types existed in 10 Vitis piasezkii Maxim. Materials.
