Isolation and expression analysis of gibberellin 20-oxidase gene from pear dwarfing Zhongai 1
To study the dwarfing mechanism of Zhongai 1 which was an excellent dwarfing rootstock bred by research institute of pomology,Chinese academy of agricultural sciences,gibberellin(GA) 20-oxidase gene was isolated from the young leaves of Zhongai 1 by reverse transcription polymerase chain reaction(RT-PCR)and rapid-amplification of cDNA end(RACE),the gene,its putative amino acid sequences and its expression intensity at different growing stages of new leaves from various pears were analyzed,the results were as following: the full-length cDNA consists of 1 179 nucleotides and encodes a putative protein of 392 amino acid residues.The putative protein has all the conserved functional active sites and typical unit of the GA20-oxidase gene family.The putative protein sequence shared 91.58% identity with that of the protein found in apple.So we confirmed that the gene was the GA20-oxidase gene of Zhongai 1,and named it as PcGA20ox1(GenBank accession numbers was HQ833589).The expression intensity of PcGA20ox1 in leaves of the three pear varieties had the same trend as Zhongai 1〈Jinxiang〈Zaosu in all tested growing periods,which were consistent with the plant height.This results indicated that the expression intensity of PcGA20ox1 was related with plant height as well.
