Genetic diversity and relationship of plum resources in Guizhou analysed by ISSR markers
[Objective]The aims of this study were to provide a scientific basis for the conservation and utilization of plum accessions in Guizhou.[Method]Genetic diversity and relationship of 45 plum accessions in Guizhou and 3 plum accessions introduced from the United States were evaluated by ISSR-PCR.[Result]The PCR by 27 ISSR primers yielded a total 198 bands and 181 scorable polymorphic markers,the number of polymorphic bands accounted for 91.41% of total bands.The Nei's genetic diversity index (H)was 0.041 2 to 0.499 2,the Shannon index of diversity(Ⅰ) was 0.102 1 to 0.693 1,and the genetic similarity coefficient among all plum accessions ranged from 0.50 to 0.93,indicating that the plum accessions in Guizhou had abundant genetic diversity.48 plum accessions could be classed into six groups at a Jaccard coefficient of 0.67,and 2 varieties of Jixinli,2 varieties of Jiuqianli,3 varieties of Yuhuangli and 4 varieties of Suli were proved to be homonyms,respectively.[Conclusion]The substantial genetic diversity was identified among the plum accessions in Guizhou,and the plum accessions of homonym could be differentiated by ISSR markers successfully.
