Generation method of prognostic regulatory networks of ceRNA related with circRNA in gastric cancer
To describe the regulatory networks of competing endogenous RNA(ceRNA)related with circRNA in gastric cancer,the expression profiles of gastric cancer circRNA,miRNA and mRNA were retrieved from GEO and TCGA databases,and differentially expressed RNAs were identified,key modules were identified using the weighted gene co expression network(WGCNA).Lasso-Cox regression analysis on key module genes was performed,while survival analysis was conduct.Differential expression analysis identified 115 circRNAs and 628 miRNAs,WGCNA constructed 15 circRNA modules and 11 mRNA modules.Lasso Cox analysis identified 4 mRNAs.A series of dysregulated circRNAs,miRNAs,and mRNAs were screened,and a ceRNA network related to circRNA in gastric cancer was constructed.The constructed network can deepen the understanding of circRNA related regulatory mechanisms.
