Effect of oxidative injury on the expression of HSPD1 gene in mouse muscle tissue
To study the effect of oxidative damage on the expression of HSPD1 gene in mouse muscle tissue,the homology of HSPD1 gene sequence in 8 species including human and mouse was analyzed using DNAman software,and the expression of HSPD1 gene after oxidative damage in different cells were queried and analyzed in GEO database.On this basis,the total antioxidant capacity(T-AOC)and the concentrations of glutathione(GSH)in mouse muscle oxidative injury models were detected,the expression of HSPD1 gene in muscle tissue was detected by qRT-PCR,and the proteins interacting with HSPD1 were predicted by online website.The results showed that the homology of HSPD1 gene sequence in 7 species including human and mouse reached 85%,and the homology of amino acid sequence was up to 97%,and the homology of amino acid sequence of HSPD1 protein between zebrafish and other 7 species was 88%.Glutathione content and total antioxidant capacity decreased after muscle oxidative injury in mice,and the expression of HSPD1 gene increased by 2.91 times,which was consistent with the changes of HSPD1 gene expression after oxidative damage in GEO database.Seven proteins interacting with HSPD1 were screened by online websites.In conclusion,oxidative damage in cells can increase the expression of HSPD1 gene,and HSPD1 protein responds to oxidative stress and oxidative damage in vivo by interacting with SIRT1 and other proteins in cells.
