Cluster Analysis of Differentially Expressed Genes of Rabbit Models of Early Knee Osteoarthritis Treated by Fumigation Therapy of Miao Ethnic Group Prescriptions
Objective:To survey the significance of hierarchical cluster analysis of differentially expressed genes(DEG)in rabbit models of early knee osteoarthritis by fumigation therapy of Miao ethnic group prescrip-tions.Methods:Forty New Zealand rabbits were randomized into four groups:blank control group,model control group,fumigation therapy group of Miao ethnic group prescriptions and fresh water control group,with ten in each group.Early KOA models were duplicated by injecting papain into the rabbit knee cavity,after successfully modeling,they were treated by fumigation therapy of Miao ethnic group prescriptions for 20 days,gene chip technology was used to perform the analysis of DEGs,consequently,to preprocess raw data obtained with RMA algorithm,and to perform cluster analysis of the gene expression spectrum.Results:The analysis of DEG showed that,when model control group was compared with blank control group,there were 827 DEGs,435 up-regulated genes and 392 down-regulated ones;when fumigation therapy group of Miao ethnic group prescriptions was compared with model control group,there were 208 DEGs,112 up-regulated ones and 96 down-regulated ones;when fresh water control group was compared with model control group,there were 152 DEGs,93 up-regulated ones and 59 down-regulated ones.Cluster analysis displayed that the samples in each experiment group were basically the same type,and each sample showed good repeatability,genetic expression of each experimental group has its own characteristics when the experiment group was compared with the control group.Conclusion:Gene chip testing could discover DEGs of fumigation therapy of Miao ethnic group prescriptions for early KOA,while cluster analysis could find the changes of the related genes and the characteristics of genetic expression.
