目的:基于网络药理学方法预测黄芩治疗慢性鼻窦炎(chronic rhinosinusitis,CRS)的有效成分及作用机制.方法:通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)收集黄芩有效成分,通过PubChem、Swiss Target Prediction数据库搜索并收集黄芩潜在靶点;通过人类基因数据库(the human gene database,GeneCards)等疾病数据库获取黄芩治疗CRS的作用靶点;运用Cytoscape软件构建黄芩治疗CRS的"药物-活性成分-疾病-靶点"网络,通过STRING数据库构建靶点蛋白-蛋白互作网络(protein-protein interactions,PPI)并进行关键靶点的基因本体(gene ontology,GO)和京都基因与基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)分析.结果:从黄芩中筛选出36个活性成分,得到黄芩与CRS共同靶点129个,涉及PI3K/Akt、RAS、Rap1、趋化因子等48条信号通路.结论:黄芩可通过多成分、多靶点、多途径发挥治疗CRS的作用.
Network Pharmacology-based Study on the Treatment of Chronic Rhinosinusitis with Huangqin
Objective:To predict the potential active ingredients and the mechanism of treating chronic rhinosinusitis(CRS)with Huangqin(Scutellaria baicalensis Georgi)using network pharmacology method.Methods:Active ingredients of Huangqin were gathered from TCMSP,the potential targets of Huangqi were searched and collected from Pubchem and Swiss Target Prediction databases;the targets of treating CRS with Huangqin were obtained from GeneCards.The network of"medicine-active ingredients-disease-targets"for the treatment of CRS with Huangqin was constructed using Cytoscape software,the protein interaction network of the targets was built via STRING database to perform biological function annotation and pathway analysis of the key targets.Results:All 36 compounds with good ADME were screened from Huangqin,it could treat CRS by intervening 129 targets and adjusting 48 pathways such as PI3K/Akt signaling pathway,RAS signaling pathway,Rap1 signaling pathway and chemokine signaling pathway.Conclusion:Huangqin could treat CRS via multiple ingredients,targets and pathways.
