Objective:To evaluate the effects of Guilong pills on the secretion of TNF-α and IL-10 in RSC96 Schwann cells induced by high glucose.Methods:After being resuscitated and cultured in a medium containing 17 mmol/L glucose for 48 h,RSC96 Schwann cells were divided into five groups:the S1 group(Vitamin D3+isotonic ethanol solution group),the S2 group(Vitamin D3+isotonic ethanol solution+high dose group of Guilong pills),the S3 group(Vitamin D3+isotonic ethanol solution+moderate dose group of Guilong pills),the S4 group(Vitamin D3+isotonic ethanol solution+low dose group of Guilong pills)and the S5 group(Vitamin D3+ethanol solution group of CYP24A1 inhibitor genistein),Vitamin D3 and the same concentration of ethanol solution were added respectively into the S1-S4 groups,ethanol solution of vitamin D3 and CYP24A1 inhibitor genistein was added to group S5 and acted for 48 hours.ELISA method was applied to detect the levels of TNF-α and IL-10 in the cell supernatant of each group in 48 hours,and the corresponding gene primer was used to measure the levels of CYP24A1m in RSC96 cells.Results:The levels of TNF-α decreased while the levels of IL-10 increased,the expressions of CYP24A1 downregulated in RSC96 Schwann cells processed with Guilong pills and CYP24A1m group.Conclusion:Guilong pills could reduce the secretion of TNF-α and increase the levels of IL-10 possibly by inhibiting the expressions of CYP24A1 protein,and indirectly regulating the levels of Vitamin D3,thereby protecting RSC96 Schwann cells induced by high glucose.
