Objective:To study the mechanism of Miao medicine Meilu Xiaocuo ointment in treating eczema,and to explore the targets of action and signaling pathways.Methods:The active ingredients-target of action of Meilu Xiaocuo ointment were searched from the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and other databases,gene name annotation was performed using the UniProt database,eczema-related targets were retrieved from OMIM,GeneCards and DrugBank.The ingredients-targets of action-pathway network of Meilu Xiaocuo ointment in the treatment of eczema was constructed using Rversion 3.6.2 software and Cytoscape 3.7.1 software,consequently,BiocManager package in R language was utilized to perform gene ontology functional annotation and KEGG pathway enrichment analysis.Results:A total of 35 active ingredients of the medicine were obtained,involving the key active ingredients such as sophoridine,quercetin,matrine,luteolin,tannin,arachidonic acid,aloe emodin;there were 97 target genes,and core proteins contained mitogen-activated protein kinase 3(MAPK3),prostaglandin endoperoxidase synthase(PTGS),mitogen-activated protein kinase 1(MAPK1),endothelial nitric oxide synthase(NOS),estrogen receptor 1(ESR1),androgen receptor(AR),brain-derived neurotrophic factor(BDNF),transcription factor and peroxisome proliferation-activated receptor(PPARG),participating in the regulation of signaling pathways such as AGE-RAGE,TNF,IL-17,MAPK and PI3K-Akt,thus playing anti-inflammatory role.Conclusion:Meilu Xiaocuo ointment could effectively relieve the symptoms of eczema,and it might be related to classic inflammatory pathways including TNF and PI3K-Akt,which could provide theoretical support for the subsequent pharmacological mechanism analysis and experimental verification of Meilu Xiaocuo ointment.
