Objective:To investigate the mechanism of glycyrrhetinic acid enhancing the chemosentivity of oxaliplatin to human colorectal cancer HT-29 cells.Methods:Colorectal cancer HT-29 cells were allocated to the control group,glycyrrhetinic acid group(the concentrations of glycyrrhetinic acid:100 μmol/L),the combination group(100 μmol/L of glycyrrhetinic acid+25 mg/L oxaliplatin)and positive control group(25 mg/L oxaliplatin).Methylthiazolyl tetrazolium(MTT)was adopted to detect the proliferation rate of HT29 cells in different groups;Annexin V/PI double labeling flow cytometry was applied to measure the apoptosis ratio of HT-29 cells in different groups;Western blotting method was used to detect the expressions of NF-κB,Bcl-2 and CASP-3 in HT-29 cells in all the groups.Results:Compared with the control group,the proliferation rate of HT-29 cells was reduced in the combination group and positive control group(P<0.05),cellular apoptosis rate was raised in the combina-tion group and positive control group(P<0.05);the relative expressions of NF-κB and Bcl-2 proteins were lowered in glycyrrhetinic acid group,the combination group and positive control group(P<0.05),while the expressions of activated CASP-3 protein were increased(P<0.05).Conclusion:Glycyrrhetinic acid could enhance the chemosentivity of oxaliplatin to human colorectal cancer HT-29 cells possibly through regulating NF-κB signaling pathway.
