Selection of Aperient Traditional Chinese Medcine Dietary FiberFormulas and Its Regulating Effect on Intestinal Flora
OBJECTIVE To study the defecation effect of traditional Chinese medcine dietary fiberformulas on mice with constipation,select the optimal formula,and explore the effect of the optimal formula on intestinal flora of mice with constipation.METHODS 6-week-old male Kunming mice were randomly divided into control group,model group,and different formula groups.A mouse constipation model was established by intragastric administration of loperamide.Each group was given corresponding drugs or solvents for intervention,and the small intestinal propulsion rate and excretion of mice in each group were observed.Time of passing the first black stool,quantity of stool,andweight of stool;collection of cecal contents,and 16S rRNAsequencing technology were used to analyze changes in intestinal flora.RESULTS Compared with the model group,the small intestinal propulsion rate and the number of black stool discharged in 5 hours in each formula groups were significantly increased.The time to discharge the first black stool in the formula 2 and formula 3 groups were significantly shortened.The weight of black feces excreted in the formula 1 and formula 2 were increased significantly at 5 hours,and formula 1 was comprehensively selected as the optimal formula.The 16S rRNA sequencing results showed that the optimal formula increased the diversity of intestinal flora in mice with constipation and changed the composition of intestinal flora.The optimal formula group increased the F/B ratio,decreased the abundance proportion of Akkermansia,and increased the relative abundance of Bacteroides and Alistipes.CONCLUSIONS The optimal dietary fiberformula can improve small intestinal motility and defecation dysfunction,adjust the intestinal flora structure and relieve constipation in mice.
