Comparative Study on Release and Transdermal Behavior in vitro between Shehuang Gels and Shehuang Powder
OBJECTIVE To compare the release and transdermal behavior in vitro between Shehuang gels and Shehuang powder.METHODS Microporous fliter membrane and isolated rat skin and advanced Franz diffusion cell were chosen in the release and transdermal behavior in vitro between Shehuang gels and Shehuang powder.The content of triphyllin A in Pronephrium triphyllum and rhein in rheum of Shehuang gels and Shehuang powder were quantitatively determined by HPLC method to determine the penetration quantity and penetration rate of the two forms.The in vitro release and transdermal results of Shehuang gels were simulation fitting by 3 equations.RESULTS The triphyllin A of Shehuang gels in 24 h were Qs=72.188 5 μg·cm-2,Q=30.458 3%,Ls=52.086 7 μg·cm-2,L=21.976 8%.The rhein of Shehuang gels in 24 h were Qs=15.220 0 μg·cm-2,Q=9.429 0%,Ls=13.545 6 μg·cm-2,L=8.391 7%.The triphyllin A of Shehuang powder in 24 h were Qs=45.239 5 μg·cm-2,Q=17.124 9%,Ls=27.043 4 μg·cm-2,L=10.237 0%.The rhein of Shehuang powder in 24 h were Qs=6.376 3 μg·cm-2,Q=1.992 1%,Ls=5.875 1 μg·cm-2,L=1.835 5%.CONCLUSION The Qs、Q、Ls and L of Shehuang gels are obviously better than the Shehuang powder.Meanwhile the Shehuang gels is less sensitized and more convenient to use in clinical.The release and transdermal model of Shehuang gels are coincides with Higuchi equation,the release and transdermal behavior of Shehuang gels are all belong to skeleton sustained-release preparations.
