Metabolism of Total Flavonoids of Desmodium Styracifolium in Rats with Calcium Oxalate Renal Stones
OBJECTIVE To explore the metabolic pathways and metabolites of total flavonoids of Desmodium styracifolium(TFDS)in rats with calcium oxalate renal stones by ultrahigh performance liquid chromatography-quadrupole-time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS).METHODS Calcium oxalate renal stones model was established in SD rats,and the rats were treated by TFDS by gavage.UPLC-Q-TOF-MS/MS was used to analyze the metabolic changes of flavone components in plasma,urine and feces of rats with calcium oxalate renal stones.The total flavone components and metabolites in plasma,urine and feces were identified by accurate relative molecular weight and multistage mass spectrometry cleavage fragments information obtained from TOF/MS and combined with the reference substance and the related literature reports.RESULTS After treated by TFDS,5 compounds were prototype constituents,3 compounds were isomers of schaftoside,and 16 components obtained by metabolism of prototype components,which were found in the plasma of rats.12 prototype components of TFDS and 21 metabolized components were identified from urine.There were 10 prototype components in feces,and 43 products were obtained by metabolism of prototype components in feces.CONCLUSION The metabolic changes of total flavonoids of Desmodium styracifolium in rats are reseached,which provides a theoretical reference for the basic research of Desmodium styracifolium in the future.
