摘要
目的 检测人参、红参、西洋参及其各部位中10种人参皂苷化合物的含量并分析评价其质量状况.方法 采用Waters ACQUITY HPLC BEH C18柱(2.1 mm× 100 mm,1.7 μm),以含0.1%甲酸溶液和乙腈-四氢呋喃(95∶5)为流动相,梯度洗脱分离,流速为0.3 mL·min-1,电喷雾正离子源(ESI+),多反应监测(MRM)模式.结果 10种人参皂苷化合物的回归曲线在各自浓度范围内线性良好,相关系数均大于0.995.平均回收率为87.7%~111.2%,RSD为1.9%~4.8%,检出限范围为0.005~0.01 μg·mL-1,定量限为0.017~0.03 μg·mL-1.结论 该法操作简便,分析速度快,灵敏度高,准确度好,适用于人参、红参、西洋参及其各部位中的多种人参皂苷化合物含量测定.同时发现,人参、红参、西洋参的须根、芦头等非药用部位含有的人参皂苷化合物总量较高,应充分利用,避免中药资源的浪费.
Abstract
OBJECTIVE To detect the contents of ten ginsenoside compounds in Panax Ginseng,red ginseng,American ginseng and their different parts,and to analyze and evaluate their quality.METHODS The chromatographic column was Waters ACQUITY HPLC BEH C18 Column(2.1 mm× 100 mm,1.7 μm).The mobile phase was composed of 0.1%formic acid and acetonitrile-tetrahydrofuran(95∶5)in gradient elution at a flow rate of 0.3 mL·min-1.A triple quadrupole mass spectrometer operating in positive electrospray ionization mode with multiple reaction monitoring was used.RESULTS The linear relationships of ten ginsenoside compounds were good in corresponding ranges with correlation coefficients(r)all above 0.995.The average recoveries of ten ginsenoside compounds were 87.7%-111.2%with the RSDs within 1.9%-4.8%.The limits of detections(LODS)for ten ginsenoside compounds ranged from 0.005-0.01 μg·mL-1.The limits of quantitation(LOQS)ranged from 0.017-0.03 μg·mL-1.CONCLUSION The method not only is convenient,rapid,sensitive and accurate,but also can be used for quantitative determination of ten ginsenoside compounds in Panax Ginseng,red ginseng,American ginseng and their different parts,Simultaneously.The total amount of ginsenoside contained in non medicinal parts such as fibrous roots and reed heads of ginseng,red ginseng and American ginseng is high,which should be fully utilized to avoid waste of traditional Chinese medicine resources.
