Correlation between the Proportion of PD-1+T-cell Subsets and Serum IFN-γ Levels in Patients with irAE
OBJECTIVE To analyze T lymphocyte subsets in peripheral blood of patients with non-small cell lung cancer(NSCLC)who developed immune-related adverse events(irAE)after treatment with immune checkpoint inhibitor(ICI).METHODS A total of 47 patients with NSCLC admitted to the First Affiliated Hospital of Guangzhou Medical University from April to October 2022 were included.Patients were divided into irAE group(n=12)and control group(n=35).Flow cytometry was used to detect the proportion of lymphocyte subsets in peripheral blood,and the difference in the proportion of lymphocyte subsets between two groups was compared.The correlation between cytokines and lymphocyte subsets was analyzed by Pearson correlation.RESULTS Compared with the control group,peripheral TCR-αβ+PD1+CD4-CD8-T cell level increased in the irAE group,and the difference was statistically significant(P=0.045).There was no significant difference in other T lymphocyte subsets.Pearson correlation analysis showed that TCR-αβ+PD1+CD4-CD8-T cells in peripheral blood were positively correlated with serum interferon-γ(IFN-γ)(r=0.448,P=0.019).CONCLUSION The level of TCR-otβ+PD1+CD4-CD8-T cell increased in the peripheral blood of irAE patients and was positively correlated with serum IFN-γ level,which may be involved in the occurrence of irAE.
