摘要

目的 利用紫外分光光度法和薄层生物自显影技术研究生脉胶囊清除ABTS自由基能力.方法 采用紫外分光光度法和薄层自显影法测定生脉胶囊中各药味清除ABTS自由基的效果,选择清除效果较强的药味进行物质分析.结果 紫外分光光度法显示生脉胶囊具有较强清除ABTS自由基能力;处方中五味子药材清除能力最强;五味子药材中木脂素类成分具有较强清除能力,其中五味子酚活性最强.结合紫外法测定结果,移植到薄层生物自显影测定法中,建立生脉胶囊清除ABTS自由基活性测定方法.薄层生物自显影法测定结果与紫外分光光度法测定结果基本一致.结论 生脉胶囊具有较强清除ABTS自由基能力,可选择薄层生物自显影技术,以五味子对照药材和五味子酚作为指标成分建立生脉胶囊清除ABTS自由基活性的测定方法.

Abstract

OBJECTIVE To determine the ABTS free radicals scavenging ability of Shengmai capsule by ultraviolet spectroscopy and thin-layer bioautography.METHODS The ABTS free radicals scavenging ability of medicinal herbs contained in Shengmai capsule was determined by ultraviolet spectroscopy and thin-layer bioautography.Those had strong antioxidant ability were selected for material analysis.RESULTS The results of ultraviolet spectroscopy showed that Shengmai capsule had the strong ABTS free radicals scavenging ability.Among the medicinal herbs contained in Shengmai capsule,Schisandra Chinensis had the strongest ABTS free radicals scavenging ability.Lignan components had the strong ABTS free radicals scavenging ability,in which,the ability of schisanhenol was strongest.Combined with the results of ultraviolet method,the ABTS free radical scavenging ability of Shengmai capsule was determined by thin-layer bioautography.The results of thin-layer bioautography were basically consistent to those of ultraviolet spectroscopy.CONCLUSION Shengmai capsule has strong scavenging ability of ABTS free radical.Using schisandra chinensis as a reference medicinal material and Schisandra phenols as indicator components,thin-layer bioautography can be usued for determining the scavenging activity of ABTS free radical in Shengmai capsules.
