摘要

目的 采用RP-HPLC-UV同时测定定喘止嗽丸中3种罂粟壳生物碱(吗啡、可待因、罂粟碱)的含量.方法 采用Kromasil100-2.5 C18色谱柱(250 mm×4.6 mm,5 µm),以乙腈-0.1%磷酸水为流动相,梯度洗脱;流速为1.0 mL·min-1;柱温:30 ℃;检测波长为210 nm(吗啡、可待因)和251 nm(罂粟碱).结果 吗啡、磷酸可待因和盐酸罂粟碱分别在0.2957~9.856 1 μg·mL-1(R2=0.9999)、0.072 9~2.429 8 μg·mL-1(R2=0.999 8)、0.046 4~1.473 0μg·mL-1(R2=0.9998)内线性关系良好;平均加样回收率为89.5%~91.5%,RSD为1.10%~3.91%.结论 该方法精密稳定,重复性好,可用于定喘止嗽丸的质量控制.

Abstract

OBJECTIVE To establish an RP-HPLC-UV method for the simultaneous contentdeter mination of morphine,codeine,papaverine from Opium Poppy Shell in Dingchuanzhisou pills.METHODS The separation was performed on a Kromasil 100-2.5 C18 column(250 mm×4.6 mm,5 μm)with the mobile phase comprising of acetonitrile-0.1%phosphoric acid flowing at 1.0 mL·min-1 in a gradient elution manner,the column temperature was 30 ℃ and the detection wave length was set at 210 nm(morphine,codeine)and 251 nm(papaverine).RESULTS Good linear relationship was observed for morphine,codeine phosphate and papaverine hydrochloride in the range of 0.295 7-9.856 1 μg·mL-1(R2=0.999 9),0.072 9-2.429 8 μg·mL-1(R2=0.999 8),and 0.046 4-1.473 0 μg·mL-1(R2=0.999 8),respectively.The average recoveries were 89.5%-91.5%with the RSD values of 1.10%-3.91%.CONCLUSION This precise stable and reproducible method can be used for the quality control of Dingchuanzhisou pills.
