Simultaneous Determination of Three Alkaloids from Opium Poppy Shell In Dingchuanzhisou Pills by HPLC
OBJECTIVE To establish an RP-HPLC-UV method for the simultaneous contentdeter mination of morphine,codeine,papaverine from Opium Poppy Shell in Dingchuanzhisou pills.METHODS The separation was performed on a Kromasil 100-2.5 C18 column(250 mm×4.6 mm,5 μm)with the mobile phase comprising of acetonitrile-0.1%phosphoric acid flowing at 1.0 mL·min-1 in a gradient elution manner,the column temperature was 30 ℃ and the detection wave length was set at 210 nm(morphine,codeine)and 251 nm(papaverine).RESULTS Good linear relationship was observed for morphine,codeine phosphate and papaverine hydrochloride in the range of 0.295 7-9.856 1 μg·mL-1(R2=0.999 9),0.072 9-2.429 8 μg·mL-1(R2=0.999 8),and 0.046 4-1.473 0 μg·mL-1(R2=0.999 8),respectively.The average recoveries were 89.5%-91.5%with the RSD values of 1.10%-3.91%.CONCLUSION This precise stable and reproducible method can be used for the quality control of Dingchuanzhisou pills.
