Study on the Relationship Between Anti-Fatigue Components and Pharmacodynamics of Traditional Southern Medicine Morinda Officinalis
OBJECTIVE To explore the correlation between anti-fatigue components of traditional southern medicine Morinda officinalis and drug efficacy,and to provide research basis for its active ingredients study and quality standard improvement.METHODS The fingerprint of anti-fatigue components of Morinda officinalis was established by HPLC.The anti-fatigue effect of the tested substance was evaluated by the mouse weight-bearing swimming experiment.The correlation between anti-fatigue components and anti-fatigue effects of Morinda officinalis from different batches was analyzed by grey correlation degree Analysis(GRA)and partial least squares regression analysis(PLSR).RESULTS The components detected from different batches of Morinda officinalis were monotropein,Deacetylasperulosidic acid,2-methylanthraquinone,methyl isoalizarin,Deacetylasperulosidic acid methyl ester,aglycoside and liquiritin.The content of monotropein was significantly higher than that of other components.GRA analysis showed that the correlations degreesof the above components and anti fatigue efficacy were 0.883 5,0.839 3,0.835 9,0.807 4,0.791 6,0.735 1 and 0.704 9,respectively.PLSR analysis showed that the components positively correlated with the anti-fatigue efficacy were Deacetylasperulosidic acid methyl ester,Deacetylasperulosidic acid,monotropein and 2-methyl-anthraquinone according to the magnitude of variable importance projection(VIP).The VIP values of the first three components were much higher than that of 2-methyl-anthraquinone and almost similar.CONCLUSION The anti-fatigue efficacy of Morinda officinarum is the result of theco-action of several components.Based on the results of grey correlation analysis and partial least squares regression analysis,the most correlated anti-fatigue effects were monotropein,Deacetylasperulosidic acid,and Deacetylasperulosidic acid methyl ester,among which monotropein was correlated and had the highest content.Therefore,monotropein can be considered as the characteristic target component of anti-fatigue activity,and the feasibility of its use as an index component for content determination should be further studied.
