Establishment of Fingerprints of Fufang Xiangru Liquid and Its Effects on SARS-CoV-2 Infected Mouse Model
OBJECTIVE To establish a quality control method for fingerprints of Fufang Xiangru liquid and to evaluate its effect on the mouse model of novel coronavirus(SARS-CoV-2)nasal drip infection.METHODS The volatile components in Fufang Xiangru liquid were analyzed by gas chromatography to establish the fingerprint of its.Mice were given Fufang Xiangru liquid 50.51 and 101.01 g crude drug·kg-1·d-1 by prophylactic gavage.On the 5th day of administration,a mouse model of viral infection was induced by nasal drops of SARS-CoV-2(4×105 PFU),and then therapeutic administration was given for 5 days,and the mice were observed for the changes in body mass,protection against mortality,and changes in lung histopathology.RESULTS The Fufang Xiangru liquid fingerprints were used to identify 11 common peaks,which were consistent with the characteristic components of the relevant herbs in the compound formula,and the constructed Fufang Xiangru liquid fingerprints had high separation,good peak shapes,many characteristic peaks,and high batch-to-batch similarity.Compared with the normal control group,the animals in the model control group showed decreased body weight,weakness and death,and lung tissues showed interstitial pneumonitis,alveolar luminal proteinaceous fluid exudation,and perivascular/peribronchiolar inflammatory cell infiltration.Compared with the model control group,the positive control group,Fufang Xiangru liquid low and high dose groups had lower mortality and significantly longer survival time,which had an ameliorating effect on the above pathological changes.CONCLUSION The fingerprint of Fufang Xiangru liquid is established,which can comprehensively and accurately evaluate the quality of the volatile components of Fufang Xiangru liquid,and Fufang Xiangru liquid has a protective effect on the viral infection model induced by SARS-CoV-2 nasal drip in mice.
