摘要
目的 探讨八珍抗癌方对结直肠癌细胞LINC01711、KLF7、Klotho的影响,并基于该通路探讨其抗癌机制.方法 选择结直肠癌细胞SW620为研究对象,以八珍抗癌方配制药液灌胃裸鼠2周后取含药血清,将细胞株分组干预后,观察不同组的LINC01711、KLF7、Klotho基因的表达情况.结果 加入含八珍抗癌方的实验组,与对照组两两比较,LINC01711、KLF7、Klotho表达差异有统计学意义(P＜0.05).其中,加入八珍抗癌方的实验组Klotho表达均高于对照组,LINC01711、KLF7表达低于对照组.另外,LINC01711基因OE组(OE代表该基因过表达)的LINC01711基因表达高于其他组,同时,KLF7基因OE组的LINC01711基因表达也高于无过达表组,此种情况同样发生于KLF7基因的表达上.结论 八珍抗癌方可以通过增加Klotho蛋白的表达来发挥抗癌作用,具体机制可能是通过抑制LINC01711/KLF7互作链的信号通路,增加Klotho蛋白的表达,来发挥抑制结直肠癌细胞的增殖的作用.
Abstract
OBJECTIVE To explore the effects of Bazhen anti-cancer formula on colorectal cancer cells LINC01711,KLF7 and Klotho and investigate its anti-cancer mechanism based on this pathway.METHODS Colorectal cancer cell line SW620 was selected as the research model the drug-containing serum was taken from nude mice after 2 weeks of oral administration of Bazhen anti-cancer formula drug solution.The cell lines were divided into groups for intervention and the expression of LINC01711 KLF7 and Klotho genes in different groups was observed.RESULTS Adding the experimental group containing Bazhen anti-cancer formula there were statistically significant differences in the expression of LINC01711,KLF7 and Klotho compared pairwise with the control group(P＜0.05).Among them the experimental group treated with Bazhen anti-cancer formula had higher Klotho expression than the control group while LINC01711 and KLF7 expression were lower than the control group.In addition the LINC01711 gene expression in the OE group(OE represents overexpression of the gene)was higher than that in other groups.At the same time the LINC01711 gene expression in the KLF7 gene OE group was also higher than that in the no overexpression group which also occurred in the expression of the KLF7 gene.CONCLUSION Bazhen anti-cancer formula exerts its anti-cancer effect by increasing Klotho protein expression.The specific mechanism may be to inhibit the signaling pathway of LINC01711/KLF7 interaction chain increase the expression of Klotho protein and exert the effect of inhibiting the proliferation of colorectal cancer cells.
