摘要

目的 探究葛根素降低LPS诱导的人单核细胞THP-1炎症反应的作用.方法 对经LPS诱导的炎症反应模型细胞添加葛根素进行处理,以Elisa检测模型细胞IL-iβ、IL-6、TNF-α分泌量差异.结果 经葛根素(浓度范围30~250 μmol·L 1)处理后,与对照组相比,THP-1模型细胞IL-1β、IL-6、TNF-α的分泌量均显著下降.结论 葛根素可显著下调人单核细胞经LPS诱导后的促炎因子分泌量,降低其炎症反应程度.

Abstract

OBJECTIVE To investigate the inhibitoryeffect of puerarin on the lipopolysaccharide(LPS)-induced inflammatory response in THP-1(human monocytes).METHODS THP-1 cells were induced by LPS to activate theinflammatory response and then treated with puerarin.After that secretion of interleukin 1β(IL-1β),IL-6,tumor necrosis factor alpha(TNF-α)were detected using the ELISA method.RESULTS The secretion of IL-1β,IL-6,and TNF-α in inflammatory THP-1 cells was significantly reduced in the puerarin-treated groups(30-250 μmol·L-1)compared to the model group.CONCLUSION Puerarin can significantly reduce LPS-induced proinflammatory cytokine secretion in inflammatory THP-1 cells,therebylessening the severity of the inflammatory response.
