Determination of 3 kinds of chromones in different parts of agarwood by biology induction
Objective To determine the chromones' contents in the agarwood generated by biology induction method and evaluate the effect of the new technology.Methods High performance liquid chromatography (HPLC) was used for the determination of aquilarone B,aquilarone C and (5S,6R,7R,8S)-2-[2-(4-methoxy-phenyl)-ethyl]-5,6,7,8-tetrahydroxy-5,6,7,8-tetrahydro-chromones (CX16) in different parts of the agarwood.Waters Symmetry Shield C18 column (4.6 mm×250 mm,5 μm) was employed eluting gradiently with acetonitrile-0.1% phosphoric acid water.The flow rate was 1 mL/min,and the detection wavelength was 234 nm.The column temperature was 30 ℃,and the injection volume was 10 μL.Results The linear ranges of aquilarone B,aquilarone C and CX 16 were 14.32-716,0.884-44.2,and 1.54-77 μμg/mL,respectively.B1 sample contained the highest contents of aquilarone B (3.411 8mg/g) and aquilarone C (1.021 6 mg/g) among the knot incense part (part B) located outside of Aquilaria sinensis xylem,and the highest content of CX16 is 0.728 5 mg/g in B4 sample.The highest contents of aquilarone B (1.424 9 mg/g) and aquilarone C (0.585 9 mg/g) among the little knot incense part (part Z) located inside ofAquilaria sinensis xylem was in Z1 sample,and the highest content of CX16 is 0.158 9 mg/g in Z4 sample among part Z.Conclusion The contents of 3 chromones varied in different parts of the agarwood induced by biological stimulation.They were significantly higher in part B than in part Z.The new method might affect the xylem with generating agarwood gradually from outside to inside.
